Prediction of psilocybin response in healthy volunteers

Besides drug dose being the most important predictor of psilocybin, other variables that followed in importance included the personality trait of absorption, performance-related activity, emotional excitability, psychological distress, drug pre-experience, and frequencies of THC and alcohol consumption. The results are limited by large proportions of unexplained variance in outcome variables and that associations between predictors and outcomes are observational. These results confirm that intra- and interindividual differences in psilocybin responses relate to differences in set and setting.
Study Summary
A meta-analysis was completed to analyze the order of importance of 24 non-pharmacological predictor variables on psilocybin response in comparison to drug dose. The analysis involved pooled data of 23 controlled experimental studies with 409 psilocybin administrations to 261 healthy volunteers. Non-pharmacological variables contributing to psilocybin response included a high score on the Absorption personality trait, being in an emotionally excitable state immediately before drug intake, and experiencing few psychological problems in the past weeks. These were strongly associated with pleasant and mystical-type experiences.
