Psilocybin with psychological support for treatment-resistant depression: an open-label feasibility study

Psilocybin appears to decrease depressive symptoms in patients with treatment-resistant depression, with these changes being maintained for up to 3 months. Future randomized controlled trials with larger sample sizes are needed to verify these results.

In an open-label feasibility trial, 12 patients with moderate-to-severe treatment-resistant depression received two oral doses of psilocybin (10mg and 25mg) separated by 7 days. Patients were assessed on measures of drug effects at each session, and were also assessed on measures if depressive symptoms at baseline, 1 week and 3 months after the second psilocybin session. A significant increase in self-reported drug effects and the impaired cognition, anxiety, and global score of the 11D-ASC were seen during the drug sessions. When compared to baseline, depressive symptoms were significantly lower at 1 week and 3 months following the 25 mg session.
