LSD modulates effective connectivity and neural adaptation mechanisms in an auditory oddball paradigm

LSD administration reduces the magnitude of the response to deviant stimuli and modulates neural adaptation mechanisms at the scalp. LSD alters both intrinsic connectivity in primary auditory areas and backward extrinsic connectivity in the fronto-temporal generative network. These results show that LSD modulates local perceptual learning mechanisms but also reduces top-down suppression of prediction error in response to auditory stimuli. The results are limited by the short inter-stimulus interval used and the small sample size.

Twenty healthy subjects underwent a pseudo-randomized, placebo-controlled, magnetoencephalography (MEG) auditory oddball paradigm study to examine the effects of LSD on feedback and feedforward connections in perceptual learning and inference under the predictive coding framework. Subjects received LSD and placebo on different days. Scalp level Event Related Fields (ERF) showed reduced neural adaptation to familiar stimuli and an attenuated neural 'surprise' response to novel stimuli in the LSD condition. The presentation of both novel stimuli and LSD modulate backward extrinsic connectivity.
