Effects of Psilocybin-Assisted Therapy on Major Depressive Disorder: A Randomized Clinical Trial

Psilocybin-assisted therapy can have rapidly effective antidepressant effects that persist one month after treatment in patients with MDD. In contrast to ketamine-like medications, psilocybin has a low risk for addiction and adverse events. These results are limited by the lack of a placebo control, a small sample size and the lack of ethnic diversity within the sample.
Study Summary
Twenty-four participants with moderate to severe Major Depressive Disorder (MDD) episodes underwent a randomized trial to test the effects of psilocybin on MDD. Patients were randomly assigned to either an immediate treatment group or a delayed treatment control group, where patients did not receive psilocybin treatment for eight weeks. Both interventions contained two psilocybin sessions, lasted 8 weeks, and had a one-month follow-up. The primary outcome measure was depression severity, measured by the GRID-Hamilton Depression Rating Scale (GRID-HAMD). Compared to the delayed treatment group, participants who received immediate psilocybin therapy exhibited decreased GRID-HAMD scores one month after treatment.
