Acute, subacute and long-term subjective effects of psilocybin in healthy humans: a pooled analysis of experimental studies

Experimental data from 227 psilocybin administrations demonstrate safety and tolerability both acutely and long-term. Acute adverse reactions and transient emotional instability are the biggest concerns when administering psilocybin. These effects were positively alleviated through strong interpersonal support and the strong control of environmental conditions. This indicates that psilocybin administration is well-tolerated in healthy subjects and future studies are warranted. Limitations include poor generalizability of the sample and sample self-selection bias.

A meta-analysis was completed to analyze the acute, short- and long-term subjective effects of psilocybin in healthy humans. Raw data was pooled from eight double-blind placebo-controlled studies conducted between 1999-2008 and included 110 healthy subjects who received 1-4 oral doses of psilocybin. Most subjects described the dose-dependent changes in mood, perception, thought and self-experience as pleasurable, enriching and non-threatening. Acute adverse drug reactions did not need psychopharmacological intervention.
