A small dose of LSD impacts amygdala connectivity. The increase in connectivity between the amygdala and the middle frontal gyrus was positively correlated with positive mood. The changes seen in amygdala connectivity are consistent with previous research using higher doses of LSD and other psychedelics. The results are limited by the use of a single LSD dose, when microdosing of LSD requires repeated doses. Additionally, the sample consisted of only healthy subjects.

Twenty healthy subjects underwent a within-subjects, double-blind, randomized trial to examine the effects of one low 'microdose' of LSD (13 micrograms) on resting-state functional connectivity and cerebral blood flow. Subjects received either the placebo or LSD dose, and participated in two fMRI scanning sessions seven days apart. Amygdala and thalamic connectivity were analyzed and mood questionnaires were collected. The LSD microdose increased amygdala seed-based connectivity with the right angular gyrus, right middle frontal gyrus and the cerebellum, while decreasing amygdala connectivity with the left and right postcentral gyrus and the superior temporal gyrus.
