Psilocybin dose-dependently causes delayed, transient headaches in healthy volunteers

Despite the dose-dependent inductive effects of psilocybin on headache, the headaches experienced by these subjects were not severe or disabling, indicating that future psilocybin experiments can be conducted safely. Psilocybin-induced headaches may possibly be due to various mechanisms. Future studies should use more detailed headache scales to differentiate between potential mechanisms. Future psilocybin experiments should also screen subjects for history of migraines.

Eighteen healthy subjects underwent a randomized, double-blind, crossover study to examine the relationship between the psychedelic, psilocybin and headache. Subjects received a range of psilocybin doses from 0, 5, 10, 20 to 30 mg/70 kg. Psilocybin frequently induced headache. The incidence, duration and severity of headache increased dose-dependently. All headaches had delayed onset, were transient and lasted no more than a day following administration.
