LSD acutely impairs working memory, executive functions, and cognitive flexibility, but not risk-based decision-making

LSD significantly impairs executive functions and working memory compared to placebo. Risk-based decision-making was unaffected by LSD. The LSD-induced cognitive deficits were normalized by ketanserin. These results implicate the role of the 5-HT2A receptor in cognitive processes of executive functions, cognitive flexibility and spatial working memory. These findings also provide a new therapeutic target for improving cognitive dysfunctions in psychiatric disorders. The results are limited by the small sample size.

Twenty-five healthy subjects underwent a double-blind, randomized placebo-controlled within-subject design to test the effects of LSD on cognition, while examining the role of the serotonin 5-HT2A receptor. Subjects received 100 micrograms of LSD alone or received LSD along with 5-HT2A receptor antagonist ketanserin. Cognitive measures included executive functions, cognitive flexibility, spatial working memory, and risk-based decision-making. LSD impaired executive functions, cognitive flexibility and working memory but did not influence the quality of decision-making and risk-taking. Ketanserin normalized all LSD-induced effects.
