Duloxetine reduces pain after Total hip arthroplasty: a prospective, randomized controlled study

Ninety-six patients scheduled to undergo a primary, unilateral total hip arthroplasty (THA) were randomized to receive daily doses of 60 mg of oral duloxetine (n=48) or placebo (starch; n=48) at 2 days pre-surgery until 14 days post-surgery. The primary outcome of interest included pain during movement on the visual analogue scale (VAS) measured at 3 hours, 6 hours, 12 hours, 1 day, 2 day, 3 days, 1 week, 3 weeks, and 3 months post-THA. Results of this trial revealed that VAS pain during movement was significantly lower in the duloxetine group vs placebo at 3 hours, 6 hours, 12 hours, 1 day, 2 days, 3 days, 1 week, and 3 weeks post-surgery (p<0.05 for all). However, no significant difference was observed for VAS pain during movement at 3 months post-surgery between the 2 groups (p=0.469).
