Exploratory Controlled Study of the Migraine-Suppressing Effects of Psilocybin

A single low oral dose of psilocybin showed significant reductions in migraine frequencies two weeks after administration. The lack of correlation between the change in migraine frequency and acute psychotropic effects suggests that the effects of psilocybin on migraines is independent of changes in sensation and perception. There were also significant reductions in weekly migraine attacks, pain severity, attack-related functional impairment and weekly migraine abortive days after psilocybin treatment. While these findings are promising, future studies are required with a larger sample size and a fully randomized design.

12 adult migraine patients underwent an exploratory, proof-of-concept, double-blind, placebo-controlled, cross-over study to test the effects of psilocybin on migraines. Patients received a placebo and psilocybin dose and outcome measures were taken two weeks after administration. The primary outcome measures were the change in migraine frequencies, change in weekly migraine attacks, light sensitivity, sound sensitivity, nausea/vomiting and attack-related functional impairment. There was a significant reduction in weekly migraine days after psilocybin treatment compared to the placebo. These changes in migraine frequency were not correlated with the intensity of acute psychotropic effects. Psilocybin was well-tolerated, with no serious adverse events.
