Collagen protein supplementation does not modulate biomarkers of bone metabolism following total hip arthroplasty: a randomized, double-blind, placebo-controlled trial.

40 patients undergoing primary total hip arthroplasty (THA) for hip osteoarthritis were analyzed after being randomized to receive hydrolysed collagen protein supplementation or an energy-matched maltodextrin placebo. The outcome of interest was procollagen type 1 N-terminal propeptide (P1NP), a marker of bone formation. Other outcomes included C-terminal telopeptide of type 1 collagen (CTX-1), inflammatory and amino acid biomarkers, pain, hip function, and health-related quality of life. Outcomes were assessed before surgery, immediately after surgery, and at 1, 4, and 15 days postoperatively, with patient-reported outcomes assessed before surgery and at days 4 and 15. Overall, the results revealed marked postoperative changes in P1NP and CTX-1, but these responses did not differ between collagen and placebo groups. Although pain at rest improved more in the collagen group, other patient-reported outcomes were similar between groups. Therefore, short-term collagen supplementation did not appear to modify early bone turnover following THA.
