Acute LSD effects on response inhibition neural networks

Findings show that 5-HT2AR activation by LSD leads to a hippocampal–prefrontal cortex-mediated breakdown of inhibitory processing, which might subsequently promote the formation of LSD-induced visual imageries. Further studies are warranted to understand the causality of this relationship. To better understand the relationship between cognitive impairments and visual hallucinations after 5-HT2AR stimulation, future studies might also want to conduct multiple tests to assess prefrontal-medicated cognitive control and reality monitoring processes.
Study Summary
In this double-blind, randomized, placebo-controlled, cross-over study, LSD (100 μg) and placebo were administered to 18 healthy subjects with a washout period of 7 days in between sessions. Response inhibition was assessed using a functional magnetic resonance imaging Go/No-Go task. LSD-induced visual hallucinations were measured using the 5 Dimensions of Altered States of Consciousness (5D-ASC) questionnaire. Results demonstrate that 5-HT2AR activation by LSD led to deficits in inhibitory processing mediated via reduced parahippocampal–prefrontal activation. LSD produced significantly higher scores than placebo for cognitive impairments and visual hallucination as indexed by impaired control and cognition.
